objective: To investigate the effective method of establishing bladder Orthotopic transplantation tumor model with human bladder cancer cell line BIU-87 to provide theoretical basis for the study of experimental animals of bladder cancer. Methods: 20 BALB-C nude mice were divided into group A and B (10 PCs), group A were injected with human bladder cancer cell line BIU-87 into the bladder wall through surgical method,B group were instilled with human bladder cancer cell line BIU-87 through bladder, progression of bladder tumor of the two groups was respectively observed in different stage . All mice were executed at experimental endpoint (week 4),tumor size and histological detection were evalued. Results: A mouse of group A died on the first day after operation, tumor occured in 8 of the rest surviving mice,tumor formation rate was 89%; no tumor occured in B group. Conclusion: Ideal orthotopic transplantation tumor model cannot established through bladder Instillation, surgery method to establish Orthotopic transplantation tumor model can definitely simulate development process of human bladder tumor disease better, which enjoy applicational value on the study of Chinese bladder tumors.
