Abstract:To investigate the effects of poly(ADP-ribose) polymerase-1(PARP-1) on cisplatin resistance of prostate cancer cells.Methods The expression of PARP-1 was down regulated by PARP-1 siRNA.The protein levels of PARP-1,Bcl-1,Bax,ERK and pERK were detected by Western blot or RT-PCR.The cell viability and IC50 of cisplatin were deteceted by the CCK-8 assay.Results The expression of PARP-1 protein was increased markedly in PC-3/DDP vs.PC-3 cells(P<0.01).Reduced PARP-1 expression could enhance the sensitivity to cisplatin in PC-3 cells, rescue the resistance of cisplatin in PC-3/DDP,and downregulate the protein levels of Bcl-2 and pERK.Conclusions Expression of the PARP-1 protein in prostate cancer cells is related to cellular sensitivity to cisplatin.PARP-1 siRNA treatment could increases the sensitivity of prostate cancer cell to cisplatin.The mechanism may be promote inhibit apoptosis pathway and phosphorylation of ERK.
